Prof. François-Clément Bidard and Dr Barbara Pistilli share expert insights on the latest TROPION-Breast01 update, presenting the final OS results from the ESMO Virtual Plenary 2025.

This study evaluates datopotamab deruxtecan (Dato-DXd) versus chemotherapy in previously treated, inoperable or metastatic HR+/HER2– breast cancer.

In this video, the experts explore:

  • Mechanism of action of Dato-DXd
  • TROPION-Breast01 PFS results and safety outcomes
  • Final overall survival results and their clinical implications
  • The future outlook for Dato-DXd

Clinical takeaways

  • TROPION-Breast01 final OS results did not reach statistical significance; however, subsequent ADC treatment may have influenced survival outcomes
  • The dual primary endpoint of PFS demonstrates a statistically significant and clinically meaningful improvement with Dato-DXd compared with chemotherapy
  • Secondary efficacy and PRO endpoints continued to show a benefit for Dato-DXd in this final analysis

  • Recognise the efficacy and safety profiles of TROP2-directed ADCs for patients with advanced or metastatic HR+/HER2- BC, and their place in the treatment landscape
  • Implement optimization of treatment selection, and make the appropriate sequencing decisions
  • Be able to monitor and manage adverse events associated with ADC therapy

François-Clément Bidard, MD PhD, is a Professor of Medical Oncology in Paris and Saint-Cloud, France. His clinical work is dedicated to breast cancer care, whereas his translational and clinical research focuses on all forms of liquid biopsy and new therapies to move ahead of cancer resistance.

Prof. François-Clément Bidard has received financial support/sponsorship for research support, consultation, or speaker fees from the following companies:

Prof. Bidart discloses financial support/sponsorship from AstraZeneca, Daiichi-Sankyo, Caris, Exact Sciences, General Electrics Healthcare, GSK, Inatherys, Lilly, Merck KGaA, Menarini Silicon Biosystems, Menarini/Stemline, Novartis, Pfizer, Prolynx, Rain Oncology, Roche, SAGA Diagnostics, Seagen, Sanofi

Barbara Pistilli, MD is the chair of the breast cancer unit at Gustave Roussy since March 2021. The clinical activity of Breast Cancer Group encloses around 2000 new patients and 700 inclusions in clinical trials, yearly. The goal of the group is to provide groundbreaking and patient-centered strategies that cover all steps from breast cancer prevention to development of innovative treatment strategies and survivorship care. The scientific strategy is driven by the integration between translational and clinical sciences in a limited number of large comprehensive programs, where knowledge in biology drives clinical development and patient samples feed translational research. In this regard, she leads a large translational program focusing on identification of mechanisms of response and resistance to antibody drug conjugates which include a number of phase I and II trials based on diverse treatment strategies in patients with metastatic breast and lung cancer, with a large sample collection for exploratory biomarker analysis.

Dr Barbara Pistilli has received financial support/sponsorship for research support, consultation, or speaker fees from the following companies:

AstraZeneca, Seagen, Gilead, Novartis, Lilly, MSD, Pierre Fabre, Daiichi Sankyo, Olema and Pfizer
